Enzymatic Digestion and HPLC Analysis of Homogenate with Pepsin and Pancreatin

This study explores the digestion of a homogenate using pepsin at pH 2.0 for 30 minutes at 37°C, followed by treatment with pancreatin and bile at pH 6.5 for 60 minutes. The effectiveness of lactase treatment, with 300 units at 37°C for 20 minutes, is also examined. Caco-2 cells are utilized for analysis, and post-digestion extracts are analyzed by HPLC. This research highlights the importance of enzymatic activity and pH in digestive processes and their role in understanding nutrient absorption.
