Testing of CCR5-specific shRNAs. Human and macaque CCR5 sequences were cloned into the psiCHECK-1 vector downstream of the Renilla translational stop codon. Initiation of the RNAi process by shRNAs that target CCR5 results in cleavage and subsequent degradation of the fusion mRNA and a reduction in luciferase expression. The vectors were co-transfected with plasmids that expressed shRNAs that target CCR5 at the indicated positions from a U6 Pol III promoter. The relative luciferase expression level is shown normalized to a control with no CCR5 target. Both CCR5-specific shRNAs target sequences in human and macaque CCR5 mRNAs at 100% identity.
